Abstract : Different molecular methods such as RT-PCR, pyrosequencing and NGS technologies were firstly tested to analyse cytochrome b from single-sporangia strains and then applied to field populations of Plasmopara viticola. RT-PCR assay design failed to analyse nucleotides insertions observed in P. viticola cytb gene, their detection relying only on pyrosequencing approach or NGS. Short reads (Illumina or Ion Torrent) and long reads (Oxford Nanopore Technologies, ONT) sequencing show that different genotypes, and consequently many resistant phenotypes to complex III inhibitors can co-exist in the same population. Interestingly, long-read ONT sequencing can detect multiple mutations/insertions along the same cytb sequence and may be used to assess potential fungicides multiple resistance in vineyard.
