Abstract : In this study, P. viticola single-sporangia strains isolated from filed populations exhibiting low sensitivity to zoxamide were characterized. Sensitivity assays on leaf discs revealed that isolated P. viticola strains exhibited high levels of resistance (EC50 >40 mg.L-1) to zoxamide compared to the sensitive strain (EC50 <0.1 mg.L-1). Two single nucleotide mutations T715A and G716C (TGC → AGC/TCC) were detected in the ß-tubulin gene and led to the C239S substitution in zoxamide-resistant P. viticola isolates. Finally, we developed a pyrosequencing method allowing the quantification of these mutated alleles, validated it on single sporangia strains and used to screen field populations. To our knowledge, it is the first report describing the molecular mechanism of resistance to zoxamide in P. viticola strains isolated from the field.
